Agile has proven itself in small teams over and over again.  When true collaboration takes hold, and communication becomes clean and transparent, work accelerates and quality usually goes up. But replicating that team esprit d'corps at the enterprise level where 1000s of developers, analysts, testers and hundreds of teams must collaborate and sync, is a another kettle of fish altogether.
